چکیده

Background Coeliac disease and IgE-mediated allergy to wheat are immune-mediated conditions which are thought to be triggered by digestion-resistant wheat proteins. Recently, a prolyl endopeptidase from Aspergillus niger (AnPEP) has been identified as being able to accelerate breakdown of gluten in food using an in vitro digestion system. Such digests have been analysed in terms of coeliac disease t-cell epitope levels[1] but have not yet been considered regarding the impact on IgE-reactivity. The amount of AnPEP required to be effective also has yet to be investigated.

Serum immunoglobulin E and immunoglobulin G reactivity to Agaricus bisporus proteins in mushroom cultivation workers

Background and Purpose: Although molds are regarded as the main fungal allergen sources, evidence indicates that spores of Basidiomycota including Agaricus bisporus (A. bisporus) can be also found at high concentrations in the environment and may cause as many respiratory allergies as molds. The aim of the present study was to evaluate specific immunoglobulin E (IgE) and immunoglobulin G (IgG) ...

A chimeric IgE that mimics IgE from patients allergic to acid-hydrolyzed wheat proteins is a novel tool for in vitro allergenicity assessment of functionalized glutens

BACKGROUND Acid-hydrolyzed wheat proteins (acid-HWPs) have been shown to provoke severe allergic reactions in Europe and Japan that are distinct from classical wheat allergies. Acid-HWPs were shown to contain neo-epitopes induced by the deamidation of gluten proteins. However, products with variable rates of deamidation can be found. OBJECTIVES In this work, we studied the effect of the exten...

Experimental assessments of the cross-reactivity of IgE from patients sensitised with acid-hydrolysed wheat protein in a cosmetic soap

Hydrolysed wheat protein (HWP), which has high emulsifying property and water retentivity, is added to various foods and cosmetics to improve their texture. Recently, several studies showed that a specific HWP formula could induce IgE-mediated hypersensitivity by skin contact and/or food ingestion. In Japan, a number of HWP-caused systemic wheat allergy cases were reported, including wheat-depe...

Fractionation and identification of the allergic proteins in Aspergillus species

Background and Purpose: Allergy is an undesired immune response to non-pathogenic agents. However, some opportunistic microorganisms such as fungi can also cause allergy. Among those fungi, hyphae form of Aspergillus strains including A. fumigatus, A. flavus, and A. niger could be mentioned. In this study, we aimed to separate allergic proteins from Aspergillus strains and determine their ident...

Prolyl endopeptidase-mediated destruction of T cell epitopes in whole gluten: chemical and immunological characterization.

Celiac Sprue is a widely prevalent immune disease of the small intestine induced by dietary gluten intake in genetically susceptible individuals. It has been suggested that prolyl endopeptidases (PEPs) may be useful catalysts for gluten detoxification. We have investigated this hypothesis using food-grade gluten as the target antigen, and a combination of mass spectrometry and patient-derived T...
